The press briefing for X06 will begin in just under one hour, (Noon Central time, 1pm Eastern, etc.) and Microsoft is hyping it up in a big way:

We’ll be showing LIVE action footage as it happens in Barcelona on Wednesday 27 September at 6pm. Get ready for some BIG announcements.

Another section on Xbox.com says:

There’s big news brewing in Barcelona. Last year in Amsterdam we unveiled the Xbox 360â„¢ launch titles. How can we beat that? We’re confident our news and announcements will blow you away, so keep your browser here for the official word first, only on Xbox.com.

Major Nelson is backstage at the show now and will be providing live text updates of the announcements on his site.  I’ve never seen him do that before, so they must have some huge announcements to drop today.  I won’t be doing live updates for the briefing, I’ll be watching it all the way through and taking note of what is said and then afterwards posting what will hopefully be a huge entry talking all about what was announced.  I’m sure whatever is announced during the conference will instantly also be uploaded onto the Xbox Live Marketplace for everyone to see right at home.  E3 lives on in X06.
